During their life, from breeding colony until the end of the experiment, laboratory animals are exposed to a variety of experiences to which they must adapt. These experiences are related to the experimental procedures applied as well as to their living conditions and both might influence the experimental outcome. In general, most investigators are aware of specific variables such as room temperature and humidity but tend to ignore the range of variables related to experimental procedures and conditions. Consequently, there\'s a need for increased awareness and control of these variables when designing a research model. Until now, few articles have been published that identified these variables and that study their impact on the welfare of the animals as well as on experimental outcome (WÏ‹rbel, 2007; Castelhano-Carlos and Baumans, 2010). In the work underlying this thesis we focused on the impact of housing- and husbandry conditions and experimental procedures in order 1) to improve the quality of experimental data by limiting confounding factors that might affect experimental data, either in a qualitative or quantitative way, and 2) to reduce the variance of experimental results. The ultimate goal of the thesis was to suggest ways by which these variables can be eliminated or their effects minimized, to improve quality of research and to allow for a reduction in the number of animals per experiment and/or refinement of the experimental protocols. The outcome of the work has generated significant new knowledge that can be used to improve research models and experimental settings. A spin-off will be an increased validity of research models and housing- and husbandry conditions, which in addition better satisfy the animals\' behavioural needs. Consequently, the project is in line with the 3R concept of Russell and Burch (1959).

Bepaalde standaard experimentele condities en procedures die behoren bij bestaande testprotocollen verhinderen behandelingseffecten zichtbaar en meetbaar te maken. Dit heeft effect op de validiteit van de onderzoeksresultaten en daarmee gevolgen voor het vertalen van deze resultaten naar o.a. de humane situatie. De bruikbaarheid van deze standaarden kan dus beperkt zijn. Vanuit de bestaande wet- en regelgeving rondom het gebruik van proefdieren is er aandacht voor de standaard experimentele condities en procedures. Nieuwe inzichten op het gebied van dierenwelzijn en de toegenomen aandacht voor dit onderwerp zijn aanleiding geweest voor aanpassingen. In de bestaande wet- en regelgeving rondom het gebruik van proefdieren wordt nu vooral nadruk gelegd op het belang van de aanwezigheid van voorzieningen voor proefdieren, die de mogelijkheid bieden om natuurlijk gedrag zoveel mogelijk te kunnen ontplooien om zo de gezondheid en het welzijn van de dieren te waarborgen en mede de validiteit van onderzoeksresultaten. Vanuit wettelijk verplicht biomedisch onderzoek is aangegeven dat deze benadering wellicht verstorend werkt op standaardisatie, omdat bestaande gestandaardiseerde testprotocollen aangepast moeten worden waardoor de herhaalbaarheid van het onderzoek op een (veronderstelde) negatieve manier beïnvloed wordt. De experimenten in dit proefschrift hebben laten zien dat geoptimaliseerde condities voor ratten en konijnen goed gestandaardiseerd kunnen worden binnen bestaande onderzoeksprotocollen zonder de variatie in experimentele resultaten te vergroten of de precisie van de proeven te reduceren, terwijl de relevantie van de testprotocollen en dierenwelzijn verbeterd werden.
